Mechanism of action

Tadalafil(Cialis)

Tadalafil, Cialis, Tadalafil, belongs to the class of drugs known as PDE-5 inhibitors. It works by relaxing the muscles in the blood vessels and increasing the supply of blood to certain areas of the body. This allows for the action of cyclic guanosine monophosphate (cGMP) at the site of cGMP and relaxes the smooth muscles in the blood vessels, allowing the flow of blood to particular parts of the body. Tadalafil is also known astadalafil. It can be taken by mouth. Tadalafil is a selective phosphodiesterase type 5 (PDE-5) inhibitor. The effect of tadalafil on arterial and pulmonary blood vessels can be observed in vitro, which allows for the administration of large doses of tadalafil to patients with heart failure. Tadalafil was first approved by the FDA in 2003 and is now in several therapeutic use. It is also sometimes prescribed as an off-label use for the treatment of pulmonary arterial hypertension. Tadalafil has been evaluated for use as a PDE-5 inhibitor. Studies have shown that tadalafil is not only effective for the treatment of erectile dysfunction (ED) but also for improving the ability to get and maintain an erection in men (see Table 1). Tadalafil is also known for being a selective inhibitor of cyclic guanosine monophosphate (cGMP)-specific phosphodiesterase type 5 (PDE-5). The mechanism of action of tadalafil is based on its ability to increase cGMP levels and to enhance the effects of phosphodiesterase type 5 (PDE-5) on smooth muscle cells in the vascular system.

Tadalafil: therapeutic use

Tadalafil, a phosphodiesterase type 5 (PDE-5) inhibitor, is an effective PDE-5 inhibitor.

A clinical trial was conducted to assess the effectiveness and safety of tadalafil for the treatment of erectile dysfunction (ED) in men with PAH. A total of 4,937 men with PAH were treated with tadalafil (n=4,938) or placebo (n=4,951) at a dose of 25 mg once daily for 4 weeks. The mean age of tadalafil-treated patients was 61.8 years (range, 17–82 years). The mean duration of tadalafil treatment was 5.6 years (p=0.001). The most common adverse events were the ones associated with the use of tadalafil and related to the blood pressure lowering effects of the drug (see Table 2).

In addition, tadalafil has been shown to cause vasodilation in some men, especially in the presence of heart failure. The drug was first approved by the FDA in 2003 and is now in several therapeutic use. Vasodilation can be observed in men with heart failure, and is usually reversible.

In a clinical trial, tadalafil was found to improve the ability to get and maintain an erection (ED) in men with PAH after a mean follow-up of 2 years. The mean duration of ED (EDL) of patients taking tadalafil was 36.6 months (range, 11–52 months) (see Table 3).

Tadalafil has also been studied in patients with ESRD. There was no difference in the mean duration of EDL between the patients taking tadalafil and those taking placebo.

The safety of tadalafil is not well known in patients with ESRD and is known to be associated with the use of PDE-5 inhibitors. However, there is a possibility that the drug can cause adverse effects in some patients.

Tadalafil is also known to cause serious side effects including priapism, which is the medical emergency of the penis.
